Biología Celular

La Unidad de Biología Celular (UBC) es una plataforma de base tecnológica cuya misión es ofrecer asesoramiento y soporte a grupos de investigación o entidades privadas en diferentes proyectos que emplean los cultivos celulares y la citometría de flujo como metodología central. Desde 2007, esta unidad lleva a cabo acciones coordinadas con el Servicio de Clasificación Celular y Citometría de Flujo (SECIF) del Instituto de Investigaciones Biológicas Clemente Estable (IIBCE).

Mediante el uso de estas tecnologías desarrollamos proyectos de investigación en el área de la biotecnología.

La UBC también integra el Programa de Tecnología Molecular, Celular y Animal (ProTeMCA), uno de los programas avalados institucionalmente.

Líneas de investigación

Tecnología celular
Durante los últimos años, nuestro grupo se ha dedicado a la generación y caracterización de líneas celulares recombinantes de interés en biomedicina y biotecnología. Entre ellas se encuentran una gran variedad de líneas celulares reporteras (IFN tipo I, NF-kB, biosensores redox, entre otros). Estas se emplean para buscar y caracterizar sustancias que interfieren con las vías de señalización de los IFN tipo I (Burgi et al, 2012 y Burgi et al, 2016), en modelos in vitro de inflamación (NF-kB, Tiscornia et al, 2012; Mastropietro et al, 2015; Rolny et al, 2016) o para mejorar el metabolismo/productividad de células de interés biotecnológico (biosensores redox en el marco del ProTeMCA).

Toxicología ambiental
Los perturbadores endócrinos (PE) son sustancias antropogénicas presentes en el ambiente, capaces de alterar la homeostasis del sistema endócrino de los organismos contribuyendo al desarrollo de patologías. Nuestra hipótesis de trabajo es que el incremento de ciertas patologías reproductivas está causado, en parte, por la creciente exposición a PEs presentes en el ambiente. En este contexto, hemos validado un modelo in vivo, el ratón transgénico Oct4-GFP, para el monitoreo toxicológico de estrógenos ambientales (Porro et al, 2015). Asimismo, hemos desarrollado un ensayo in vitro que emplea una línea celular reportera dual y permite evaluar en un único test la actividad estrogénica o androgénica de un PE putativo. En esta línea de investigación estamos colaborando con el Dr. Rodríguez (ISAL, CONICET-UNL, Argentina).

Péptidos antitumorales
Desde el 2011 colaboramos con la Dra Vallespi, Centro de Ingeniería Genética y Biotecnología (CIGB), Cuba, en el proyecto “CIGB-552: péptido novedoso con propiedades antitumorales y anti-inflamatorias útiles para el tratamiento del cáncer”. Demostramos que el CIGB-552 es eficaz para reducir el tamaño de tumores presentes en ratones e identificamos a la proteína COMMD1 como mediador clave para su actividad antitumoral (Fernández Massó et al, 2013, Vallespí et al., 2014, Núñez de Villavicencio et al, 2015). Recientemente, describimos la unidad funcional mínima del CIGB-552 necesaria para ejercer su actividad biológica (capacidad de penetrar células tumorales, interactuar con COMMD1 e inducir apoptosis, Astrada et al, 2016 y 2018).

Ingeniería de tejidos: organoides intestinales y cultivos 3D
Los cultivos celulares tradicionales, en dos dimensiones, ofrecen un sistema de evaluación simple, rápido, económico y reproducible. Sin embargo, carecen de la capacidad de recrear las interacciones celulares que tienen lugar en un tejido in vivo, limitando su poder predictivo. Nuestro objetivo consiste en generar y caracterizar modelos celulares en tres dimensiones (3D) que asemejen a los sistemas vivos, mejorando la correlación de los resultados. En particular, nos proponemos establecer cultivo de organoides intestinales murinos, ya sea a partir del tejido primario o mediante la diferenciación de células madres pluripotentes inducidas (del inglés iPSC) y aplicarlos en el estudio de la inflamación, el cáncer y el cribado de probióticos.

  • Disponemos de varios laboratorios equipados para trabajar en un ambiente aséptico, como cabinas de seguridad biológica, incubadoras de CO2, centrífugas citológicas y microscopios invertidos. Además contamos con un Analizador Automático para Glucosa y Lactato Bioprofile Basic 2 Analyzer (Nova Biomedical).
