Bioinformática

La bioinformática es la aplicación de técnicas computacionales y estadísticas para el análisis datos de origen biológico, que incluye también su organización y almacenamiento así como el desarrollo de herramientas para manejarlos.

El volumen de datos generado actualmente por las técnicas de secuenciación promueve un desarrollo constante de métodos para lidiar de manera eficiente con numerosos genomas/proteomas, sin perder de vista los datos biológicamente relevantes.

En nuestra Unidad aplicamos variadas herramientas al estudio comparativo de genomas a escala global, así como familias específicas de genes y proteínas. Históricamente nos hemos focalizado en bacterias, patógenos humanos y el genoma humano. También hemos desarrollado herramientas específicas para lidiar con los nuevos desafíos metodológicos planteados por la explosión de información. Estas aplicaciones y desarrollos tienen cabida en genética y biomedicina, y en la más reciente metagenómica, la caracterización genómica de muestras ambientales, pudiendo ser el ambiente un curso de agua o la mejilla interna de un individuo.

Líneas de investigación

Genómica humana
El proyecto URUGENOMES busca crear capacidades locales en el área de genómica humana y en especial genómica médica. En este contexto, se secuenciaron 50 genomas, los que están siendo analizados desde el punto de vista de genómica poblacional (ancestrías), y otros 30 genomas de pacientes con enfermedades raras, los cuales se analizan en el contexto de genómica médica.

Transcriptómica humana
Una de las áreas en que la unidad se especializa es la transcriptómica humana. Dentro de esta línea se están desarrollando actualmente varios proyectos. Uno de ellos, por ejemplo, se dedica al estudio de procesos de diferenciación de células madre a diferentes tipos celulares (adipogénesis, osteogénesis y cardiomiogénesis). De especial interés es la regulación post transcriptional que tiene lugar durante estos procesos. Otro de los proyectos dentro de la línea tiene como objetivo el análisis de la respuesta de células humanas frente a la infección de patógenos de distintos orígenes.

Bioinformática estructural
En el contexto de nuestra Unidad se busca explotar datos de estructuras de proteínas, generados por cristalografía de rayos X y otras técnicas (RMN, crio-electromicroscopía), en la comprensión funcional de familias de proteínas. En un ida y vuelta entre estructuras tridimensionales y secuencias genómicas, este ejercicio provee muchas veces pistas sobre la función de una proteína y, a veces, genera hipótesis funcionales sobre familias enteras de genes ‘hipotéticos’ (genes sin función conocida).

Genómica y evolución microbiana
Busca comprender la evolución de patógenos que afectan al ganado y las mascotas utilizando la filodinámica; y fundamentalmente en el descubrimiento de las fuerzas evolutivas que dan forma a los genomas de bacterias zoonóticas como Campylobacter, Leptospira y Mycobacterium. Entre los proyectos en curso dentro de esta línea se destaca un esfuerzo conjunto de América Latina para estudiar la dinámica de la población de Clostridium difficile utilizando enfoques de epidemiología genómica. Recientemente, uno de los proyecto de esta línea busca analizar la dinámica de resistencia a antibióticos en enterobacterias de entornos urbanos mediante la aplicación de metagenómica.
