Cell Biology
The Cell Biology Unit (UBC) is a technology-based platform whose mission is to offer advice and support to several research groups or industries in different projects that use cell cultures and flow cytometry as a main technology. Since 2007, our unit carries out collaborative actions with the Servicio de Clasificación Celular y Citometría de Flujo (SECIF) from Clemente Estable Institute.
Using these technologies (cell culture and flow cytometry) we develop research projects in the area of biotechnology.
The UBC is also member of the Molecular, Cellular and Animal Technology Program (ProTeMCA), one of the institutionally supported programs.
Members
Research lines
1) Cellular Technology: Over the past 20 years, our group has focused on the generation and characterization of recombinant cell lines of interest in biomedicine and biotechnology. Among these are a wide range of reporter cell lines, including NF-kB, Type I IFN, redox biosensors, and others. These are used to identify substances that interfere with signaling pathways, to develop in vitro models, or to study inflammatory processes. To stay at the forefront of knowledge, we have launched a new line of work involving the establishment of intestinal organoids from animal species to study infectious/zoonotic diseases. These models, in addition to being versatile tools for developing complex high-content experiments, contribute to the reduction of animal use in research, making us pioneers in Uruguay and the region in this field.
2) Flow Cytometry: Our group is a national and regional leader in flow cytometry and has developed innovative methods and new tools to address biological questions through conventional cytometry techniques. In 2023, we installed the first full-spectrum cytometer in Uruguay, significantly expanding our analytical and resolution capabilities, with the added ability to subtract autofluorescence in samples where necessary. To remain at the cutting edge of knowledge and enhance human resource development, we organized and delivered an international course in November 2023.
3) Antitumor Peptides: For 15 years, we have collaborated with Dr. Vallespi from the Center for Genetic Engineering and Biotechnology (CIGB) in Cuba on the project “CIGB-552: A New Peptide with Antitumor Properties for Cancer Treatment.” We demonstrated that CIGB-552 is effective in reducing tumor size in mice and identified the COMMD1 protein as a key mediator of its antitumor activity. For the first time, we showed that the CIGB-552 peptide contains the minimal sequence necessary to produce the most significant biological effects, which are mediated by its ability to penetrate cells through transduction and endocytosis mechanisms. The activity is primarily linked to the predominant internalization mechanism, and to a lesser extent, to the abundance of the COMMD1 protein. Furthermore, CIGB-552’s ability to negatively modulate NF-kB and HIF-1 pathways is reduced in the COMMD1 knockout cell line, validating its role in the cytotoxic mechanism. As a result of this successful collaboration, we were honored with the 2023 National Academy of Sciences Award from Cuba.
